Preparation of microbial cells: Safety precautions; sterility; types of growth media (liquid, solid); identification and classification of microorganisms: morphology, Gram stain, biochemical tests; measurement of microbial concentrations; comparison of growth rate in differently constituted media; selective action of antibiotics. Cell fractionation: Cytoplasm, Nuclear, Mitochondria, Mitoplast; Assay for enzyme markers e.g. succinate dehydrogenase. Use of protein assay to monitor cell growth: Cell growth; cell density; centrifugation; protein determination. Cell Behaviour: Cell interactions; kinetics of cell pairing; phagocytosis; kinetics of phagocytosis; exocytosis. DNA technology: Isolation, purification and manipulation of DNA; RFLP, PCR, Hybridization.
